• BMI of 27 kg/m² or above with a weight-related condition, such as high blood pressure, prediabetes, type 2 diabetes, abnormal blood fats, obstructive sleep apnoea or certain cardiovascular conditions

Treatment should be used alongside a reduced-calorie diet and increased physical activity. The suitability of the Foundayo pill will be assessed individually by a clinician before it is prescribed.

How effective is the Foundayo pill for weight loss?

The Foundayo pill was studied in the Phase 3 ATTAIN-1 trial, involving 3,127 adults with obesity or overweight and at least one related health condition, without diabetes. Participants took orforglipron or placebo once daily for 72 weeks alongside dietary changes and increased physical activity.

Using the efficacy estimand, participants receiving the highest 36 mg investigational dose achieved an average body weight reduction of 12.4% after 72 weeks. At this dose:

• 77.1% lost at least 5% of their body weight
• 59.6% lost 10% or more
• 39.6% lost at least 15%
• 20.1% lost 20% or more

The study also reported improvements in cardiometabolic measures, including waist circumference, systolic blood pressure, triglycerides and non-HDL cholesterol.

Important: These results are based on the efficacy estimand from the ATTAIN-1 trial and do not indicate how much weight an individual will lose. Results with orforglipron vary depending on individual factors and response to treatment.

What Foundayo dosages are available?

There are six Foundayo dosage strengths, starting at 0.8 mg once daily. The dose is increased gradually, with each step usually lasting at least 30 days to give your body time to adapt.

You may not need to reach the maximum Foundayo dosage. Our clinician will consider how you are responding and tolerating treatment before recommending any increase. Always follow your prescribed dosing plan and only change your dose when advised by our clinician.

How do you take Foundayo?

Take Foundayo once daily at a time that suits your routine, with or without food. Swallow the tablet whole and do not crush, split or chew it. Do not take more than one tablet within 24 hours. There is no need to fast or plan your dose around meals. Taking Foundayo at a similar time each day may make it easier to remember. What happens if you miss a Foundayo Dose?

If you forget your dose, take the tablet when you remember, provided you have not already taken Foundayo that day. Never take two tablets at once to make up for a missed dose.

What should you do if you take too much Foundayo?

Contact our clinician promptly if you take more Foundayo pill than prescribed. Taking too much may increase the likelihood of side effects, including nausea and vomiting.

What happens when you stop Foundayo?

Which Foundayo side effects can occur?

As with other prescription medicines, Foundayo side effects are possible, but they do not affect everyone. Gastrointestinal effects, including nausea, diarrhoea and vomiting, are reported most frequently and may be more noticeable when beginning orforglipron treatment.

• Low blood sugar when Foundayo pill is used without insulin or a sulfonylurea

Foundayo warnings and safety advice 

Understanding Foundayo warnings can help ensure treatment is used safely. Before prescribing, our clinician will consider your medical history, existing conditions and any factors that could affect whether the medicine is appropriate for you. Do not take Foundayo tablets if you have an allergy to orforglipron or any other ingredient in the medicine. Before treatment, let our clinician know if you have or have previously experienced:

• Pancreatitis
• Diabetes treated with insulin or sulfonylureas, which can increase the risk of low blood sugar
• Gastroparesis or other severe digestive problems
• Kidney problems, particularly alongside vomiting or diarrhoea
• Low blood pressure or treatment for hypertension
• Gallbladder problems
• Diabetic eye conditions, including retinopathy or macular oedema

Does Foundayo interact with other medicines?

Before taking the Foundayo pill, tell our clinician about any prescription medicines, over-the-counter treatments, supplements or herbal products you use. Some treatments can change the amount of orforglipron in your body, while Foundayo tablets may affect how certain other medicines work. Medicines that can increase orforglipron levels include:

• Clarithromycin
• Itraconazole or ketoconazole
• Ritonavir or telaprevir

• St John’s wort

Can Foundayo affect birth control?

The orforglipron pill may reduce the effectiveness of some oral hormonal contraceptives. If you take the contraceptive pill, let our clinician know before starting treatment.You may be advised to use an additional barrier method, such as condoms, or switch to a non-oral contraceptive. This may be needed for 30 days after starting the Foundayo tablet and for 30 days after each dose increase.
